摘要

目的 调查12 个X 染色体STR 基因座在浙江汉族人群的遗传多态性,为法医学应用提供基础数据。方法 应用ZJGA‐X12 荧光标记复合扩增系统,对浙江汉族468名无关男性个体与449 名无关女性个体进行 DXS7133 、DXS8378 、DXS981 、DXS7424 、DXS6789 、DXS10159 、GATA165B12 、DXS101 、DXS7423 、GATA31E08、DXS10164 、DXS10162 这12 个X‐STR 基因座的复合扩增,用ABI3130XL 型基因分析仪对扩增产物进行检测,并统计这12 个X‐STR 基因座的群体遗传学参数。结果 获得12 个X‐STR 基因座的等位基因频率分布,分别检出8 、7 、13 、12 、11 、8 、7、16 、6 、8 、9 、11 个等位基因,获得男性样本DXS10159‐DXS10162‐DXS10164与DXS101‐DXS7424 两组连锁基因座单倍型119、62 种;分别统计了12 个X‐STR 基因座的GD 、DP 、MEC 等法医遗传学参数。结论 12个X‐STR 基因座具有较强个体识别能力,可应用于法庭科学中的个体识别与亲权鉴定。

Abstract

 Objective  To study the genetic polymorphism of12 X‐short tandem repeat (X‐STR) loci of Han population in Zhejiang province , and evaluate their forensic application . Method  DXS7133 ,DXS8378 ,DXS981 ,DXS7424 ,DXS6789 , DXS10159 ,GATA165B12 ,DXS101,DXS7423 ,GATA31E08 ,DXS10164 and DXS10162 loci were amplified by using ZJGA‐X12 PCR amplification Kit ,following analyzed by 3130XL genetic analyzer .468 unrelated male individuals and449unrelated female individuals of Han population in Zhejiang province were investigated to determine the distributions of allele frequencies .Results   Allele distributions of the12X‐STR loci have been obtained ,8 ,7 ,13 ,12 ,11,8 ,7 ,16 ,6 ,8 ,9,11 alleles were found in the preview 12 X‐STR loci ,119 and 62 haplotypes of two pairwise linkage groups of DXS10159‐DXS10162‐DXS10164 and DXS101‐DXS7424 were found in male samples .The combined discrimination power(DP) ,mean exclusion chance(MEC) and gene diversity(GD) of 12 loci is calculated .Conclusion  The 12 X‐STR loci are highly polymorphic and suitable for forensic personal identification and paternity testing .
