本文旨在构建基于国产测序平台的遗传标记复合检测体系,提升法医 DNA 鉴定在个体识别和亲缘关 系分析中的技术能力。基于华大 G99RS 高通量测序平台,筛选 48 个常染色体 STR、32 个 X-STR、65 个 Y-STR 基因座以及线粒体 DNA 高变区(HV-Ⅰ、HV-Ⅱ、HV-Ⅲ)构建检测体系,从重复性、灵敏度、种属特异性、 抗抑制能力、混合样本解析能力及与毛细管电泳一致性等方面进行系统验证。结果显示,1 ng DNA 模板量 STR 和 Mt-SNP 平均测序深度分别为 5 285× 和 26 765×,平均覆盖度分别为 86.66% 和 86.48%。体系在批次 内和批次间均表现出良好的重复性;灵敏度可达 62.5 pg;对 9 种常见动物 DNA 表现出优异的种属特异性;在 腐殖酸(≤ 200 ng/μL)、单宁酸(≤ 225 ng/μL)、血红素(≤ 1 000 μmol/L)和靛蓝(≤ 3 000 μmol/L)存 在下保持稳定的扩增效能;与毛细管电泳分型结果的一致性达 99.86%。本研究成功构建了基于国产高通量测 序平台的遗传标记复合检测体系,为法庭科学深入研究提供更多解决方案,为案件的侦破提供更多线索。
