Analysis of Lorazepam in Blood by Liquid-liquid Extraction

Abstract
Objective To isolate with liquid-liquid extraction into the lorazepam that is to analyze through gas/liquid chromatography-mass spectrometer (GC-MS/LC-MS) so that the comparison can be conducted into the difference between the two instrumental analyses of lorazepam, thus having rendered a method to detect lorazepam in blood.Methods Lorazepam-spiked sampling blood (1.00mL) was disposed with ethyl acetate (3.00mL) under vibration, afterwards centrifuged to separate the organic phase that was firstly dried at 45℃ and then dissolved with 0.1mL methanol. The dissolved substance (lorazepam) was analyzed into GC-MS/LC-MS, with its retention time, characteristic ions and relative abundance ratio being qualitatively determined plus the peak area of characteristic ions being quantified.Results For the sampling blood containing lorazepam of 1.00µg/mL, no responsiveness was turned up from GC-MS, yet LC-MS was revealing the lorazepam recoveries of 80%~103% when the lorazepam was even lowered to 50.0ng/mL.Conclusions The gas chromatography-mass spectrometry is not suitable for detection of lorazepam in blood. Reversely, the liquid-liquid extraction coupling with liquid chromatography-mass spectrometry is eligible for the purpose, demonstrating its simple, rapid and sensitive merits in tackling into the relative cases of robbery and/or rape.
