Objective The technical performance of homemade DNATyperTM Y24 kit was tested and its forensic applicability was evaluated. Methods DNATyperTM Y24 kit was verified in terms of genotyping requirements (e.g., amplification setup, primer dosage, consumption of heat-activated enzyme, Mg2+ concentration, annealing temperature, cycle numbers) and technical indicators (precision, accuracy, peak height balance, stability, species specificity, sensitivity, adaptability for various samples and mixture). Results The appropriate amplification conditions for the DNATyperTM Y24 kit are 10μL amplification volume, 1×primer, annealing temperature of 59 ℃ and 28 cycles, thus leading the kit to the accurate and stable genotyping of good repeatability and highly specific for human DNA. The lowest quantity is 0.125ng DNA for the kit to obtain complete STR genotyping. The kit is adaptable for various samples and consistent with different batches of its products. Conclusions DNATyperTM Y24 kit is suitable for application into criminal cases and DNA databasing in forensic practice.
