Objective To establish a method for evaluating the ATD-GC-MS analysis results of gasoline residues in fire debris samples. Methods Simulated-burning samples were analyzed with ATD-GC-MS, the extracting characteristic ions chromatogram of aromatics, alkanes, indane and naphthalene were compared with chromatogram of gasoline, and then the similar-degree of the fingerprint chromatogram of fire debris samples and the gasoline using vectorial angle method were calculated. Results Generally, the similar-degree were larger than 90% for the fire debris samples which with gasoline as accelerant, less than 60% for the fire debris samples which without gasoline as accelerant. Conclusion The objective, reliable and accurate evaluation can be made for the ATD-GC-MS analysis results by using the comparison of the four characteristic ions chromatogram of fire debris samples with chromatogram of gasoline, and with the similar-degree calculation of the chromatograms of the fire debris samples and gasoline at the same time.

Objective To compare the extraction effects of touch DNA by Chelex-100, DNA IQ and EQ magnetic beads. Methods The standard DNA diluted to 10ng, 100ng was treated respectively by Chelex-100, DNA IQ system and EQ magnetic beads. DNA was extracted from 30 cigarette butts and 30 toothbrushes respectively by the three methods. The quality and quantity of DNA were detected between PCR quantification and STR analysis. Results There wasn't any loss of DNA with the Chelex method, but with DNA IQ method and the EQ magnetic beads method, some DNA was lost. For cigarette butts and toothbrushes samples, yield of touch DNA and IPC CT extracted by Chelex method are higher than that extracted by DNA IQ method and the EQ magnetic beads method, but they aren't significantly different by DNA IQ method and EQ magnetic beads method. Conclusion The Chelex method is very suitable for extracting DNA from the less pollution and low impurity samples, and the DNA IQ system and the EQ magnetic beads method are more suitable for minute and polluted samples.

Objective To explore the capture and separation technology of sperm cells and to establish a reagent system for sperm cell-specific separation and DNA extraction methods respectively. Methods The sperm samples were prepared with specific capture and separation system and cell were separation from cell mixtures. Results With the help of above complexes, sperm cells were isolated from different mixtures of sperm cells and buccal cells. The specificity of reagent system was additionally checked by DNA genotyping. A complete STR genotyping was obtained by the current reagent system for the capture and separation sperm cells from cell mixtures.

Objective To present a analysis method of monofluroacetate in human blood with pre-column derivatization and liquid chromatography and ion trap electro-spray mass spectrometry. Methods The human blood samples were wiped off protein with acetonitrile, the supernatant was devivated in 60 ℃water for 90 min using α-Br-acetophenone as derivative reagent and tetrabutyl ammonium bromide as activator, the derivative was filtered through a 0.45μm membrane, and then was directly analyzed by liquid chromatography and ion trap electro-spray mass spectrometry. Results The calibration curve showed good linearities in rang of 0.15μg/mL~15.40μg/mL and the limit of detection was 0.020μg/mL.Conclusion The Results showed this method is easy to operation and has good sensitivity, it can be widely applied in the identification of monofluroacetate for casework.

Objective To explore the effects of latent fingerprint development light on subsequent DNA analysis of sweat. Methods Three groups of fingerprint were exposed to ultraviolet light for ten minutes, thirty minutes and one hour, respectively, and one group of fingerprint is non-exposed to ultraviolet light, then the four groups fingerprint were conducted DNA analysis and the Results were compared. Results The ultraviolet impairs DNA in sweat latent fingerprint. The longer time a fingerprint was exposed to ultraviolet light, the less DNA was obtained. Conclusion For development of sweat latent fingerprint with ultraviolet light, the exposure time should be as short as possible (in ten minutes).

Objective To investigate the analysis method of disperses dyes on polyester fibers by high performance liquid chromatography- mass spectrometry. Methods With DMF as the extraction agent, disperse dyes on 12 kinds fiber samples were analyzed in the negative ion mode MS.Results According to the first order mass spectrum, eight kinds of samples can be discriminated. A quasi-molecular ion with strong response signal was selected as parent ion from the ionized products of dyes, after two mass spectrometry full-scans, the rest samples were identified by the number of characteristic fragment peaks and mass-to-charge ratio in LSIMS.Conclusion The high performance liquid chromatography -mass spectrometry is an effective method for the analysis of dyes on the fiber.

According to calculation formula of vehicle speed, two methods for measuring the vehicle's speed based on the surveillance video were suggested. The first method is based on the combination of video image and field measurement. Matched with the surveillance video image, the vehicles' movement distance can be measured on field, and the movement time can be calculated from the video frame rate. The second method uses the measurement of vehicles technical parameters. While the vehicles' typical part passes the same point in the video image, the movement distance and time can be obtained from the vehicles' technical parameters and video frame rate. Two methods have been evaluated by experiments.
