Objective To obtain a single genotype of a suspect to facilitate database search, the DNA mixtures in a homicide case were de-convolved. Methods Mixtures component de-convolution using graph theory and expert analysis methods were used to analyze the mixture, the deduced genotypes were searched in the National DNA database. Results The same convicted was matched by either method. Conclusion DNA mixtures de-convolution followed by DNA Database search is a new approach in casework.

Objective To develop a method for simultaneous determination of methanol, ethanol and propanol in blood by Gas Chromatography. Methods Conditions of gas chromatography were optimized and then blood serum samples were analyzed quantitatively by GC/FID. Results Linear relations of ethanol were Y=0.4145X+ 0.0232(R2=0.9974)for the concentration ranged from 1 to 100mg/100mL and Y=0.4511X+0.0746(R2=0.9911)for the concentration ranged from 100 to 1000mg/100mL. Linear relation of methanol was Y=0.2778x+0.0493(R2=0.9983)for the concentration ranged from 1 to 200mg/100mL. Conclusion This method can be applied in clinical diagnosis of poisoning and forensic analysis of alcohol in blood.

Objective To investigate DNA analysis and DNA quantification of samples treated with the agents of benzidine tests. Methods Bloods of ten peoples were collected, and bloodstains were made on the filter paper. The same size and amount bloodstains were made by BSD600-DUET Paper Hole Puncher. Six experimental groups are divided by samples for 30min, 1h, 3h, 6h, 12h and 24h after samples treated with benzidine. Template DNA of samples were extracted by three DNA extraction methods(magnetic bead-based DNA extraction protocol,QIAamp DNA Investigator Kit protocol and Chelex-100 extraction).STR profiling was detected by PCR. DNA quantification of samples treatment was detected by real time PCR. Results The average values of DNA quantification of samples were decreased continuously with times after treatment with benzidine. The linear curves is Y=-0.4087ln(x)+0.7044(R2=0.7633)for the group of magnetic bead-based DNA extraction protocol),Y=-0.2393ln(x)+0.4764(R2=0.8715)for the group of QIAamp DNA Investigator Kit protocol, Y=-0.1178ln(x)+0.2302(R2=0.9571)for the group of Chelex-100 extraction. Statistical comparisons between each of the groups were made by multiple-ways Analysis of variance with SPSS 19.0 indicated that benzidine can alter DNA amount over time up to 24h(P<0.01), and that DNA amount with different DNA extraction methods have significant differences(P<0.01). DNA typing of 16 locis of 8.33% samples was detected by PCR-STR with Identifiler plus kits and the loss of 232 heterozygosity were detected in 142 samples. Conclusion Benzidine test has significant influence on DNA typing and DNA quantification, so the samples treated by benzidine could not be detected by PCR-STR.

Objective To research injury mechanism of Rolando fracture. Methods 49 cases clinical data of Rolando fracture and 21 experimental samples of first metacarpal bones of cadaveric hands struck with blunt force were analyzed. Results Imaging showed Rolando were T or Y shaped fracture, and fracture went through the carpometacarpal joint, and the base of the first metacarpal bone was divided into three pieces, thumb was shortened and displaced. Rolando fracture was caused by longitudinal and blunt force which struck the first metacarpal bone head and conducted along the body of metacarpal bone when thumb bending. Conclusion Rolando fracture was caused by longitudinal and indirect force.

In this paper, printing defects, or imperfections, found on counterfeit currency were examined by graphic scanning and image processing, and then the characteristics of printing defects were analyzed and classified. The results demonstrated that it is possible to associate counterfeit currencies to a common origin by analysis of printing defects.

Objective To introduce a new method for detection of latent fingerprints on thermal paper. Methods More than 300 fingerprints, including new and old fingerprints on thermal paper were developed with DFO and DFO /PVP respectively, and the results were compared. Results The fingerprints developed by using Modified DFO with PVP have better contrast. Conclusion The modified DFO method has the advantages of time-saving, higher sensitivity and stronger fluorescent.

Objective To establish a method for determining Valone in liver. Methods Valone in liver was extracted in diatomite column with codichloromethane and ether respectively, and then the extraction was analyzed by high performance liquid chromatography. Results The extraction rate were 98.1% and 99.9%, the limits of detection were 14ng/mL and 13ng/mL. Conclusion The method is simple, rapid, high extraction rate and can be used as a conventional method of forensic toxicology.
