    Objective Through initial screening of common black ink mark, provide the basis for further examination. Methods 19 kinds of black ink mark were analyzed using confocal miscroscopic laser Raman Spectrometer. Results According to the Raman spectra, 19 kinds of black ink were divided into three categories, that is no Raman peak black ink, carbon-containing black ink and dye-containing black ink .Conclusions Raman Spectrometer can be used to distinguish different kinds of black ink, which is useful for choosing suitable method to further examination.

    Objective To analyze the limitation of scoring mechanism used in automatic fingerprint identification system and explore a weighted method based on fingerprint feature point distribution. Methods A large group of fingerprints (12 million people) were studied and distribution regularity of fingerprint feature points was summarized. According to the density distribution of fingerprint feature points, the weighted factors were added into the score. Results The test matching was conducted and the results demonstrated that the weighted score of same fingerprint compared to other fingerprint was improved. Conclusions This weighted scoring mechanism based on fingerprint feature point distribution is effective for accuracy improvement of automatic fingerprint identification system.

    Objective This paper investigated the enhancement of STR profiles peak intensity using post-PCR purification, as it becomes increasingly important to gain DNA profile information from exceedingly trace levels of DNA. Methods The 9947 genome of 25pg/μl concentration was used as DNA template for STR profiling. The PCR product was divided into control group and three examined groups. The control group was electrophoresed as standard procedure, and in examined groups, PCR products were purified using molecule sieve method (Qiagen DyeEX2.0 spin kit),ultrafiltration method (Amicon Ultra-0.5,100KD) and affinity chromatography method (Qiagen MinElute Column) separately before electrophoresis. Results In three examined groups, an approximately 4-fold increase in STR peak height (RFU) was observed relative to the standard conditions, while there is no significant peak height difference between three groups. Conclusions Post-PCR purification is an alternative tool for enhancing STR peak height and would be used in forensic casework.

    Objective To establish a method for isolating spermatozoa from epithelial cells in male-female mixtures by fluorescence in situ hybridization and laser capture microdissection. Methods Male sperm and female vaginal epithelial cells from two volunteers were collected and mixed with each other, CEP?X SpectrumOrangeTM Y SpectrumGreenTMkits was used to hybridize the mixtures, and then the male and female cells were collected selectively by laser capture microdissection. Finally, a multiplex PCR was performed by the Identifiler kit using the on-chip LV-PCR. The cells found to contain only a single orange(X)or a single green(Y) were classed as spermatozoa, where as cells found to contain both an orange and a green were considered as epithelial cells. Results The completed STR typing was obtained from 20 sperm cells. The more cells were analyzed, the higher recovery rates and lower allelic dropout rates were got. With 30 sperm cells, the highest recovery rate (95%) was obtained. Conclusions The combination of Fluorescence in situ hybridization and laser capture microdissection is effective for genotyping sperm from sperm/epithelial cell mixtures.

    Objective To explore a method for qualitative analysis of benzodiazepine drugs with Raman Spectroscopy. Methods Ten types of benzodiazepine drugs were tested by using laser Raman Spectrometer and the Raman spectra were analyzed. Results Raman peak shifts of ten types of Benzodiazepine drugs are different, so all Benzodiazepine drugs can be determined by Raman Spectroscopy. Conclusions Laser Raman spectroscopy is a good way to analyze solid samples of benzodiazepine drugs.

    Objective In this study, Micro-Raman Spectroscopy was used to analysis printing inkblots. Raman spectra of 3 brands, 21 models printing inkblots were recorded with exciting of 514nm laser. According to the characteristic peaks of different Raman spectra, we can quickly distinguish different kinds of printing inkblots. It provides one reference method for printing document examination.

    Objective To evaluate the impact of psychological contamination in polygraph examination and explore the corrective and preventive measures. Methods The causes of psychological contaminations of examinee were analyzed and the psychological contaminations were classified. Based on the classification of psychological contamination, improved methods of forming question and evaluating principle were established. Results The improved methods were used in casework and the satisfactory results were achieved. Conclusions Impact of psychological contaminations of examinee can be corrected by using right methods of polygraph test.

    Objective To analyze the handwriting of blue gel ink pen by using SERS based on silver colloid. Methods 4 brands of blue gel ink pen were collected from market and the handwriting samples on paper were made with them. The concentrated silver colloid was dropped on the surface of handwriting stroke, and then the prepared samples were analyzed by Laser Confocal Raman Spectroscopy. Results Handwritings written by 4 brands of blue gel ink pen have different SERS spectrum. Conclusions Handwritings of blue gel ink pens on paper can be identified by SERS based on silver colloid.

    Objective To establish a non-deconstructive method to lift footwear impressions in urine with the plane scanning technology. Methods Footwear impressions deposited on marble, plastic flooring and ceramic tile surface with urine as a contaminant were lift by three kinds of plane scanning methods, and the angle of incident light and reflected light was adjusted to get satisfactory images. Results The contrast between the footwear impressions and background was enhanced significantly. The interference of background was reduced. Conclusions Plane scanning technique can be used to lift footwear impressions in urine as a non- deconstructive method.

    Objective To develop a rapid and accurate method for analyzing synthetic cannabinoids AKB 48. Methods Synthetic cannabinoid K3 sample was tested by gas chromatography/mass spectrometry. The temperate of injection port was 280 ℃, initial temperature of column was 80 ℃, carrier gas flow rate was 2.0 ml/min, mass spectrum sampling rate was 2. Results The chemical component AKB 48 was determined in the Synthetic cannabinoid K3 sample. Conclusions This method has the advantages of rapid, accurate and sensitive, and can be used for AKB 48 qualitative analysis.

    Objective To introduce a new method for latent fingerprints developing by using NaYF4:Er,Yb phosphor suspension,and explore its applicable scope and conditions. Methods The sebaceous fingerprints on non-porous surfaces, including plastic, tile, glass, metal et al. were developed with NaYF4:Er,Yb phosphor suspension. Results The sebaceous fingerprints on non-porous objects with complex background were developed successfully. The effectiveness of NaYF4:Er,Yb was compared with that of traditional Zinc Oxide powder, the results indicated that some benefit of NaYF4:Er,Yb over Zinc Oxide powder for the development of latent fingerprints. Conclusions NaYF4:Er,Yb phosphor suspension is a simple and effective method for fingerprints detection, and has the great potential for development of latent fingerprints.

    Objective The psbA-trnH sequence of cpDNA was utilized as a novel technique to discriminate drug original plants of cannabis and adulterants. Methods The psbA-trnH regions were amplified and sequenced bi-directionally. Then the obtained sequences were assembled using the CodonCode Aligner. The psbA-trnH genetic distances of the regions were computed in accordance with the kimura 2-parameter (K2P) model and Neighbor-joining (NJ) phylogenetic trees were constructed using MEGA5.1.Results The psbA-trnH sequences analysis showed that K2P distances of inter-and intra-specific were different significantly. The NJ trees showed that the cannabis could be distinguished easily from adulterants. Conclusions The psbA-trnH fragment could be used as a candidate barcode for identifying cannabis from adulterants and it provide a new method to accurately identify cannabis. DNA barcoding has broad prospects for plant identification in criminal investigation.

    Objective To establish a method of automatic extraction of sperms DNA from mixed stain. Methods 56 mixed stain samples were collected from casework. The samples were extracted by using DNA IQTM System and Biomek?2000 Laboratory Automation Workstation, and then analyzed with 3130xl?genetic analyzer. Results STR genetic typing was obtained from all 56 mixed stain samples. Conclusions DNA IQTM System and Biomek?2000 Laboratory Automation Workstation can be used for sperms DNA extraction from mixed stain in forensic casework.

    Objective To investigate the polymorphism of 18 STR loci in Han population from Daqing district. Methods DNATyperTM19 kit was used to detect the allelic frequencies of 18 STR loci in 221 unrelated Daqing Han individuals, and then the population genetics parameters were calculate by using statistic software. Results The results demonstrated that all loci were found to be no deviation from Hardy-Weinberg equilibrium (P>0.05), Heterozygosity (H) ranged from 0.608 to 0.896, matching probability (Pm) ranged from 0.016 to 0.196, power of discrimination (DP) ranged from 0.804 to 0.984, polymorphism information content (PIC) ranged from 0.560 to 0.910, power of exclusion (PE) ranged from 0.301 to 0.788. Conclusions The 18 STR loci are relatively abundant in polymorphic information and suitable for forensic personal identification and paternity testing.
