    Objective To deveplope a guidline for samples collection of Y-STR database in Zhuji region of Zhejiang Province. Methods 5903 male bloodstain samples were collected from 56 surnames that belong to 156 villages in 17 towns of Zhuji area. The samples were typed using a 17 Y-STRs Y-filer PCR amplification Kit. The distribution of Y-STR haplotype was analyzed according to the different combinations of town-village-surname.Results A total of 1987 different haplotypes were found in 235 combinations of town-village-surname. The number of combinations of town-village-surname-haplotype was 2547. The haplotype numbers occurred in the different combinations of town-village-surname ranged from 1 to 18. 1686 unique combination of town-village-surname-haplotype was observed. Most of haplotypes(95.3%) belong to 1-2 combinations of town-village-surname. One to two major haplotypes was detected in 90.7% town-village-surname. The minor haplotypes frequency reached 18.22%.Conclusion The Results show that more samples are need to be collected from the surnames which villages have a high ratio of minor haplytpes based on the family infromation. This may be helpful to avoid dropping some hapltypes in Y-STR database of Zhuji region in Zhejiang Province.

    In this article, we analyzed 25 samples of stamp inks and their stamp impressions on paper by HPLC, the Results showed that the chromatography difference of ink paste and its stamp impression was little, but for oil-based ink, water-based ink and their stamp impression, the chromatography difference was significant.

    Objective The aim of this study was to investigate mutation of 15 STR loci. Methods 2722 bloodstain samples collected from 817 parentage confirmed cases were detected by using IdentifilerTM system and the mutation rates of STR loci were calculated.Results With 33060 allele transfers,25 mutations were identified at 11 loci. The average mutation rates is 0.8×10-3per locus(95%CI 0.5-1.1×10-3).20 mutation events were one-step mutation, 3 events was two-step mutation, 2 events was three-step mutation; The ratio of paternal versus maternal mutations was 2.6:1, and 7 indistinguishable mutations were observed.Conclusion STR mutation were common in paternity testing, and we should attach importance to it.

    Objective To investigate the origin of Han population male family in Shouguang district of Shandong Province and to study the genetic stability of Y-chromosomal STR loci of the same population.Methods 624 unrelated male family from Shouguang district were samples, 17 Y-STRs loci were amplified and analyzed by 3130XL genetic analyzer, and the population genetic parameters were calculated.Results During about 700 years heredity and mutation accumulation, the Y-STR haplotypes of 624 unrelated Han population male family still displayed the family constancy.Conclusion Y-STR haplotypes can be used for inferring the suspect's family origin.

    Objective To explore the forensic application of Ligase Chain Reaction (LCR).Methods The human Amelogenin locus was selected for LCR. According to the sequences of M55418 on the human Amelogenin chromosome loci Xp22.1-3, and M55419 on Yp11.2, the primers was designed and the LCR system was set up for genotyping male and female.Results The genotypes of male and female Amelogenin loci can be get by nested LCR correctly.Conclusion LCR can be applied to human Amelogenin locus genotyping.

    Objective To investigate the effects of forensic UV light sources on the subsequent polymerase chain reaction (PCR)-short tandem repeat (STR) analysis of blood fingerprints and latent fingerprints.Methods Blood fingerprints and latent fingerprints on six different materials were exposed for up to 15 minutes to two different UV light sources, they were subsequently analyzed using a PCR system.Results It was found that UV light had no appreciable effect on subsequent PCR analysis for blood fingerprints but had an effect on subsequent PCR analysis for latent fingerprints.Conclusion The shorter wave and more powerful of the UV light, the longer time and shorter distance was exposed,the less DNA was obtained.

    Objective To establish a method for analysis of Cyanide in biological samples by Iso-nicotinic Acid-pyrazdone color class method.Methods The Cyanide was extracted by steam distillation. In the buffer solution at pH6.8-7.0, the Cyanide reacts with chloramine-t forming Chlorocyanogen, and then the Chlorocyanogen reacts with Iso-nicotinic and Acid-pyrazdone to form a stable blue complex.Results The content range of Cyanide was estimate by compared the color class of standards and samples.Conclusion This method is simple and accurate and can used for analysis of Cyanide in biological samples.
